I'd like to load a piece of [unknown] software on my PC so that when I select various instruments from within PowerTab for playback, that's what I get. I'm running XP.

For example, I'd like to play back my PowerTab stuff through something that's sounds like a guitar vs. a calliope. I don't know squat about what I should be looking for to do this--any recommendations?

If you have a good sound card you can re reroute the signal to an external midi sound module. In general midi guitars suck very few are realistic for more than a couple notes. I have found a couple of good sound fonts that make them less painful. But , you need a soundblaster live or some other sounhfont capable card to do that.
